The PHLPP isoforms, which stand for PH domain and Leucine-rich repeat Protein Phosphatases, encompass a pair of protein phosphatases, namely PHLPP1 and PHLPP2. These phosphatases play a crucial role in regulating Akt serine-threonine kinases (Akt1, Akt2, Akt3) as well as conventional/novel protein kinase C (PKC) isoforms. In several types of cancer, PHLPP is believed to function as a tumor suppressor due to its capability to impede growth factor-induced signaling within cancer cells. Specifically, PHLPP dephosphorylates Ser-473, which is part of the hydrophobic motif in Akt, resulting in partial inactivation of this kinase.
