YieldReaction ConditionsOperation in experiment
86% With tin(ll) chloride; In ethanol; at 80℃; for 3h; step 2: A mixture of 246 (500 mg, 2.0 mmol) and SnCl2 (2.26 g, 10.0 mmol) in EtOH (10 mL) was heated at 80 °C for 3 h. After cooling to RT, the reaction mixture was filtered, and the filtrate concentrated in vacuo. Water (100 mL) was added to the residue, followed by addition of solid NaHC03. The mixture was then extracted with EtOAc (3 x 50 mL), washed with brine (50 mL), and concentrated in vacuo to afford 440 mg of a yellow solid. Formic acid (10 mL) was added to the solid and the mixture stirred at 100 °C for 2 h then concentrated in vacuo. Water (100 mL) was added to the residue followed by addition of solid NaHC03. The mixture was extracted with EtOAc (3 x 50 mL), and the combined extracts were dried (Na2S04), filtered and concentrated to afford 400 mg (86percent) of 5-bromo-6-chloro-lH- benzo[d] imidazole (248) as yellow solid: MS (ESI) m/z = 231.1 (M+l).
