Laduviglusib (CHIR-99021) trihydrochloride serves as a potent and selective inhibitor of GSK-3α/β, with IC50 values of 10 nM and 6.7 nM. It exhibits >500-fold selectivity for GSK-3 over CDC2, ERK2, and other protein kinases. Additionally, Laduviglusib trihydrochloride acts as a potent activator of the Wnt/β-catenin signaling pathway, enhances self-renewal in mouse and human embryonic stem cells, and induces autophagy.
