Biological Activity

In Vitro:

Cell Line
Concentration Treated Time Description References
CD3+ T cells 200 µM 24 h Mito-TEMPO significantly reduced TIM3 expression in eicosenoate-treated CD3+ T cells and restored IFN-γ expression in CD4+ and CD8+ T cells, indicating that Mito-TEMPO can restore eicosenoate-induced T-cell dysfunction. PMC9981933
neonatal rat cardiomyocytes (NRCMs) 10 μM 24 h Mito-TEMPO significantly decreased PA-induced total ROS level and negated PA-induced mitochondrial ROS production, whereas apocynin had no effect on this. PMC5948205
N87-shNDUFS1 cells 10 μM 1 h Mito-TEMPO significantly inhibited the upregulation of HIF1α expression and transcriptional activity induced by NDUFS1 knockdown, further indicating that NDUFS1 promotes the activation of the HIF1α signaling pathway by inducing mROS production. PMC10575981
Immortalized mouse kupffer cells (ImKCs) 50 µM 6 h Mito-TEMPO suppressed LPS-induced mtROS production, showing similar antioxidant effects as ZC. PMC10672841
Hep3B cells 50 µM 24 h Mito-TEMPO suppressed TNF-α-induced mtROS production, showing similar antioxidant effects as ZC. PMC10672841
Primary hepatocytes 50 µM 6 h Mito-TEMPO suppressed TNF-α-induced mtROS production, showing similar antioxidant effects as ZC. PMC10672841
MSCs 100 μM 48 h For initial in vitro screening to find the candidate drug molecule which enhances mitophagy by restoring depolarized mitochondria colocalisation with the LC3-autophagosomes. PMC10181927
Bone marrow-derived macrophages 500 µM 15 min To investigate the effect of Mito-TEMPO on ATP-induced NLRP3 inflammasome activation. Results showed that Mito-TEMPO inhibited ATP-mediated IL-1β and IL-18 secretion in BMDMs from WT mice but failed to inhibit IL-1β and IL-18 secretion in CypD-KO BMDMs. PMC8477875
mouse platelets 10 μM 30 min Mito-TEMPO significantly inhibited thrombin-induced ROS generation without affecting CRP-induced ROS production. PMC10415391
AML cells 100 µM Used to detect mitochondrial reactive oxygen species (mt-ROS) levels, showing increased mt-ROS signal intensity in AML cells. PMC10470505

Dissolving Methods
Please choose the appropriate dissolution scheme according to your animal administration guide. For the following dissolution schemes, clear stock solution should be prepared according to in vitro experiments, and then cosolvent should be added in turn:

in order to ensure the reliability of the experimental results, the clarified stock solution can be properly preserved according to the storage conditions; The working fluid for in vivo experiment is recommended to be prepared now and used on the same day;

The percentage shown in front of the following solvent refers to the volume ratio of the solvent in the final solution; If precipitation or precipitation occurs in the preparation process, it can be assisted by heating and/or ultrasound.
Protocol 1
Protocol 2
The prepared working fluid is recommended to be prepared now and used up as soon as possible in a short period of time. The percentage shown in front of the following solvent refers to the volume ratio of the solvent in the final solution; If precipitation or precipitation occurs in the preparation process, it can be assisted by heating and/or ultrasound.
