YieldReaction ConditionsOperation in experiment
94% Step 3. 3,3-dimethyl-2,3-dihydro-1 H-indoleTo a solution of 1-acetyl-3,3-dimethyl-2,3-dihydro-1 H-indole (22.0 g, 1 15.8 mmol) in MeOH (100 mL) was added 4M HCI in MeOH (100 mL) and the mixture stirred at 50 C for 16 h. The solvent was then removed under reduced pressure. Water was added to the residue, the pH was adjusted to pH 8 and the aqueous layer was extracted with EtOAc. The organic layer was then dried (Na2S04), filtered and then concentrated to give 3,3- dimethyl-2,3-dihydro-1 H-indole (16.0 g, 94%). 1H NMR (400 MHz, CHLOROFORM-d) delta ppm 1 .30 (s, 6 H) 3.30 (s, 2 H) 6.62 - 6.66 (m, 1 H) 6.71 - 6.76 (m, 1 H) 7.02 (s, 2 H)

YieldReaction ConditionsOperation in experiment
97.1% With sodium hydrogencarbonate; In dichloromethane; at 0 - 20℃; To a suspension of NaHC03 (5.41 g, 64.39 mmol) and [1914-02-9]3,3-dimethylindoline (0.79 g, 5.37 mmol) in DCM (10 mL) was added acetyl chloride (0.76 mL, 10.73 mmol) dropwise at 0 C. The reaction was stirred at rt for 2 h, then filtered. The filtrate was concentrated in vacuo to give the title compound as a white solid (0.99 g, 97.1%).
