85% With Nonafluorobutanesulfonyl fluoride; 1,8-diazabicyclo[5.4.0]undec-7-ene In tetrahydrofuran for 24h; Inert atmosphere; 4 Example 4: In a nitrogen atmosphere, add 20.0g of compound 4 chiral N-tert-butyloxycarbonyl-3,4-dihydroxypiperidine (92.05mmol, 1.0eq.), 14.01g of DBU (92.05mmol, 1.0eq.) and 60g of tetrahydrofuran to the reaction bottle. Cool to 0, add 27.81g of perfluorobutylsulfonyl fluoride (92.05mmol, 1.0eq.) in tetrahydrofuran (40g) dropwise, and keep warm for 24h. After the reaction is completed, filter and concentrate, and column chromatography directly obtains 9.08g (41.42mmol) of compound 5 chiral N-tert-butyloxycarbonyl-3-hydroxy-4-fluoropiperidine mixture crude product, light yellow liquid (yield: 45%, purity: 96%, e.e. ≥ 90%). Example 5:Add 50 mL of toluene and 17.17 g of D-dibenzoyltartaric acid (42.9 mmol, 1.05 eq.) to a reaction flask, heat to 50°C, dropwise add 10.0 g of a toluene (50 mL) solution of a chiral N-tert-butyloxycarbonyl-3-hydroxy-4-fluoropiperidine mixture (45.6 mmol, 1.0 eq.) of compound 5, stir for 1 h, slowly cool to -10 to 10°C, and filter to obtain 9.44 g of compound 6 N-tert-butyloxycarbonyl-(3S, 4R)-3-hydroxy-4-fluoropiperidine as a white solid (yield: 85%, purity: 100%, e.e.: 100%).
