NS8593 hydrochloride is an effective selective SK channels (small conductance calcium-activated potassium channels) inhibitor. NS8593 hydrochloride reversibly inhibits SK3-mediated currents with a Kd of 77 nM. NS8593 hydrochloride inhibits all SK1-3 subtypes of Ca2+ dependence (Kd values of 0.42, 0.60, and 0.73 µM at 0.5 µM Ca2+, respectively) and does not affect intermediate and large conductance calcium-activated potassium channels (hIK and hBK channels). NS8593 is also an effective inhibitor of TRPM7 channels with an IC50 of 1.6 µM.
