PA-8 is an effective, selective, orally active PACAP type I (PAC1) receptor antagonist. It inhibits PACAP-induced CREB phosphorylation in PAC1 receptors but does not inhibit VPAC1 or VPAC2 receptors. PA-8 also inhibits PACAP-induced cAMP elevation (IC50 = 2 nM) and suppresses PACAP-induced aversive responses and mechanical allodynia in vivo.
