75% With ammonia; hydrogen In ethanol at 20℃; for 72 h; Ethyl l-(aminomethyl)cyclobutanecarboxylate; A solution of compound ethyl 1-cyanocyclobutanecarboxylate (2 g, 13 mmol) in 1percent ammonia in ethanol (20 mL) was hydrogenated with 5percent rhodium on alumina (0.8 g) over the weekend (3 d). The reaction mixture was filtered through celite. The filtrate was concentrated and dried to give the product, compound 3 (1.5 g, 75percent) as light yellow liquid. 1H NMR (400 MHz, CHLOROFORM-^) δ 1.29 (t, j=7.1 Hz, 3 H) 1.93-1.96 (m, 2 H) 2.33 - 2.49 (m, 4 H) 3.07 (s, 2 H) 4.18 (q, J=7.2 Hz, 2 H).
72% With hydrogen In ethanol at 40℃; for 18 h; 1-Cyano-cyclobutanecarboxylic acid ethyl ester (2.0Og, 13.0mmol) was dissolved in ethanol (30ml) and treated with Raney Nickel (1 ml as a slurry in water) and hydrogen gas at 4 bar and heating at 400C for 18 h. The reaction mixture was filtered through Dicalite and washed through with ethanol (50ml). The solvent was removed under reduced pressure to give 1-aminomethyl-cyclobutanecarboxylic acid ethyl ester as a yellow oil (1.47g, 72percent).
