CAY10526 is a specific microsomal prostaglandin E2 synthase 1 (mPGES1) inhibitor. It selectively regulates mPGES1 expression, inhibiting PGE2 production without affecting COX-2. In melanoma xenograft models, CAY10526 inhibits tumor growth, increases apoptosis, lowers levels of anti-apoptotic proteins BCL-2 and BCL-XL, and increases pro-apoptotic proteins BAX, BAK, and cleaved caspase-3. It inhibits cell viability in mPGES1-expressing melanoma cell lines (IC50 < 5 μM).
