Stage #1: dimidium bromide With sodium tetrahydroborate In methanol Inert atmosphere; Cooling with ice; Stage #2: With acetic acid In methanol Inert atmosphere; Cooling with ice; 2.5. Synthesis of hydrodimidine and 2-chlorodimidium triflluoroacetate Hydrodimidine (HD) (Fig. 1) was synthesized by reduction of dimidiumbromide (DiBr) using NaBH4, as described above for the synthesis of HE from EtBr, using HPLC to verify complete reduction (Fig. 8A). Theidentity and purity of HD was confirmed by HPLC (Fig. 8A), MS(Fig. 8B), and NMR (Table 4 & Supplementary Fig. 13-17) as describedbelow. 2-Chlorodimidium (2-Cl-D+) (Fig. 1) was synthesized by chlorinationof HD using NCS as described above for the synthesis of 2-Cl-E+from HE, and then purified as described above for the purification of 2-Cl-E+. The identity and purity of 2-Cl-D+ was confirmed by HPLC(Fig. 8A), MS (Fig. 8B), and NMR (Table 5 & Supplementary Fig. 18-24)as described below.
