Members of the FKBP family, such as FKBP12, exhibit PPIase activity. These immunophilin ligands demonstrate varying effectiveness in animal models for conditions like Parkinson's disease, dementia, and spinal cord injury. In these scenarios, canonical immunophilins act as chaperones, influencing protein folding and modulating oxidative stress. Conversely, noncanonical FKBP members like FKBP38 lack a conserved FK506-binding site, displaying neither PPIase activity nor an affinity for FK506. Intriguingly, inhibiting the noncanonical FKBP family member through small molecules seems to confer neuronal protection and stimulate the proliferation of neuronal stem cells in a rat focal cerebral ischemia model.
