97% With tetrakis(triphenylphosphine) palladium(0); potassium carbonate; In 1,4-dioxane; water; at 80℃;Inert atmosphere; A flask was charged with <strong>[24078-12-4]4-bromo-2-methylbenzaldehyde</strong> (5.00 g, 25.1 mmol, 1.00 equiv), phenylboronic acid (3.98 g, 32.6 mmol, 1.30 equiv), tetrakis(triphenylphosphine)palladium (2.90 g, 2.51 mmol, 0.10 equiv), potassium carbonate (10.4 g, 75.3 mmol, 3.00 equiv), 1,4-dioxane (200 mL), and water (40 mL) under nitrogen. The resulting solution was stirred overnight at 80 C and then quenched with water (250 mL). The resulting solution was extracted with DCM (3 x 250 mL) and the organic layers were combined, washed with brine (2 x 150 mL), dried over anhydrous sodium sulfate, filtered and concentrated under reduced pressure. The residue was (1153) chromatographed on a silica gel column to provide 4.79 g (97% yield) of 3-methyl-[l, l'-biphenyl]- 4-carbaldehyde as a colorless oil. LCMS (ESI, m/z): 197 [M+H]+.
91% With tetrakis(triphenylphosphine) palladium(0); potassium carbonate; In 1,4-dioxane; water; at 90℃;Inert atmosphere; A round-bottom flask was charged with <strong>[24078-12-4]4-bromo-2-methylbenzaldehyde</strong> (8.00 g, 40.4 mmol, 1.00 equiv), 1,4-dioxane/water (100/20 mL), phenylboronic acid (7.39 g, 60.6 mmol, 1.50 equiv), potassium carbonate (16.7 g, 121 mmol, 3.00 equiv), and tetrakis(triphenylphosphine)palladium (4.67 g, 4.04 mmol, 0.10 equiv) under nitrogen. The reaction mixture was stirred overnight at 90 C and diluted with water (100 mL). The resulting solution was extracted with ethyl acetate (3 x 100 mL) and the organic layers were combined, washed with brine (2 x 100 mL), dried over anhydrous sodium sulfate, filtered and concentrated under reduced pressure. The residue was chromatographed on a silica gel column to provide 7.19 g (91% yield) of 3 -methyl-[ 1,1 ?-biphenyl]-4-carbaldehyde.
