84.45% With porcine pancreatic lipasetype II; In aq. buffer; at 20.9℃; for 0.0633333h;pH 9.0;Enzymatic reaction;Kinetics; NH2N-Ac-Phe-OEt (50 mM) with various amounts of <strong>[4985-46-0]L-tyrosinamide</strong> and porcine pancreatic lipase were mixed with 80 mM pH 9.0 Tris-HCl buffer in screw-capped tubes. The reaction mixture was thenplaced in an orbital sharking bath (150 rpm) at designed reactiontemperature for the times shown in Table 1. The reaction of porcinepancreatic lipase catalyzed synthesis of N-Ac-Phe-Tyr-NH2is rep-resented in Scheme 1. After reaction, a 4-fold volume of terminationreagent containing ACN and acetic acid was added into the reac-tion mixture to deactivate the enzyme. Twenty microliters of the reaction mixture was injected into a high-performance liquid chro-matography (HPLC) for composition analysis.
