YieldReaction ConditionsOperation in experiment
85% With hexamethylenetetramine; at 100℃; for 4h; A mixture of 5-methoxy-1H-pyrrolo[2,3-clpyridine (From step 2, 12.5 g, 84.37mmol) and hexamethylenetetramine (17.74 g, 126.55 mmol) in HOAc (200 mL) was stirredat 100 C for 4 hours. Water (300 mL) was added to the reaction mixture and the solution wasextracted with EtOAc (300 mL x 2). The combined organic layers were dried over anhydrousNa2504 and concentrated. The residue was purified by chromatography on silica gel (0 - 4 %MeOH in DCM) to afford the title compound (12.6 g, yield 85 %) as a white solid. ?H NMR(400 MHz, CD3OD) 9.89 (s, 1H), 8.37 (s, 1H), 8.26 (s, 1H), 7.45 (s, 1H), 3.93 (s, 3H).
