YieldReaction ConditionsOperation in experiment
200 mg With piperidine; In pyridine; at 100℃; for 5h;Inert atmosphere; The starting pyrazine derivative (X04, 500 mg, 1.0 eq.) and malonic acid (481.3 mg, 1.0 eq.) were dissolved in 3.5 mL of pyridine, 3 drops of piperidine were added, and the mixture was heated at 100 °C under nitrogen for 5 hours. The reaction was monitored by LCMS and TLC. The reaction solution was diluted with water, the pH was adjusted to 5 with 1 M HC1, and then extracted twice with ethyl acetate (2X1 OmL). The organic layers were combined, dried with anhydrous sodium sulfate, filtered and concentrated to obtain the crude product A04, which was then purified by column chromatography ( EtOAc:Petroleum=l : 1) to get pure A04 (200 mg).
