Recombinant Human IL-6 is a human-derived interleukin-6 protein expressed in Escherichia coli and produced as a tag-free protein. Belonging to the IL6/GCSF/MGF family, it is a glycoprotein consisting of approximately 170–180 amino acid residues, stabilized by two disulfide bonds that maintain its globular structure, with the fourth α-helix being critical for its biological activity. IL-6 is an important pro-inflammatory and immunomodulatory cytokine expressed by various cell types, capable of inhibiting the growth of early-stage melanoma cells while promoting the proliferation of advanced-stage cells.

Biological Activity

Description
Cytokines of the IL6/GCSF/MGF family are glycoproteins of about 170 to 180 amino acid residues that contain four conserved cysteine residues involved in two disulfide bonds. They have a compact, globular fold (similar to other interleukins), stabilized by the 2 disulfide bonds. One half of the structure is dominated by a 4 alpha-helix bundle with a left-handed twist; the helices are anti-parallel, with 2 overhand connections, which fall into a 2-stranded anti-parallel beta-sheet. The fourth alpha helix is important to the biological activity of the molecule. Interleukin-6 (IL-6) is an important proinflammatory and immunoregulatory cytokine expressed by various cells. Interleukin-6 has been shown to inhibit the growth of early stage and to promote the proliferation of advanced stage melanoma cells in vitro.

Formulation Description Lyophilized from a 0.2 μm filtered solution of 20mM PB, 5% Sucorse, 4% Mannitol, 0.05% PS-80, pH7.4.
Reconstitution Method Always centrifuge tubes before opening.Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100μg/ml. Dissolve the lyophilized protein in distilled water. Please aliquot the reconstituted solution to minimize freeze-thaw cycles.
For long-term storage, it is recommended to dilute the product further with a solution containing carrier proteins (e.g., 0.1% BSA, 10% FBS, or 5% HSA). For serum-free cultures or in vivo animal experiments where BSA, FBS, or HSA cannot be present in the cytokine preparation, trehalose may be used as an alternative carrier. Aliquots should be stored frozen at -20°C to -80°C.
Note: Low-adsorption tubes are recommended for aliquot storage, and repeated freeze-thaw cycles should be avoided. The dilution concentration for aliquoting should not be lower than 10 μg/mL, and the aliquot volume should be no less than 10 μL.
Aliquoted protein remains stable for up to 3 months when stored at ≤ -20°C.
