Biological Activity

Description
AZD3839 free base is a potent and selective orally active BACE1 inhibitor that penetrates the brain (Ki=26 nM). It demonstrates a 14-fold selectivity against BACE2 and >1000-fold selectivity against cathepsin D. AZD3839 free base effectively reduces plasma, brain, and cerebrospinal fluid Aβ levels in mouse, guinea pig, and non-human primate models in a dose- and time-dependent manner. It holds promise for Alzheimer's disease research[1][2].

In Vitro:

Cell Line
Concentration Treated Time Description References
N2A cells 32.2 nmol/liter 16 hours To evaluate the inhibitory effect of AZD3839 on Aβ40 levels in N2A cells, showing that AZD3839 effectively reduced Aβ40 levels. PMC3510823
SH-SY5Y cells 4.8 nmol/liter 16 hours Promoted Nrf2 nuclear translocation and upregulated Nrf2 downstream gene expression, thereby protecting cells from oxidative stress damage PMC3510823
Guinea pig primary cortical neurons 24.8 nmol/liter Overnight To evaluate the inhibitory effect of AZD3839 on Aβ40 levels in guinea pig primary cortical neurons, showing that AZD3839 effectively reduced Aβ40 levels. PMC3510823
Mouse primary cortical neurons 50.9 nmol/liter Overnight To evaluate the subcellular localization and stress-induced nuclear TDP-43 foci formation of acetylation-mimic TDP-43K145Q in neurons. Results showed that TDP-43K145Q formed more nuclear TDP-43-positive foci under oxidative stress. PMC3510823

In Vivo:

Animal Model
Administration Dosage Frequency Description References
Mouse C57BL/6 mice Oral gavage 80 and 160 μmol/kg (35 and 69 mg/kg) single dose or 100 μmol/kg (43 mg/kg) repeated doses Single dose or twice daily for 7 days To evaluate the inhibitory effect of AZD3839 on plasma and brain Aβ levels in mice, showing that AZD3839 reduced plasma and brain Aβ levels in a dose- and time-dependent manner. PMC3510823
Mice J20 transgenic AD mouse model Intraperitoneal injection 200 mmol/kg or 100 mmol/kg Single dose or twice daily for 7 days AZD3839 significantly rescued the abnormal early electrophysiological phenotype of J20 mice and reduced the accumulation of b-CTF fragments. PMC5952042

Dissolving Methods
Please choose the appropriate dissolution scheme according to your animal administration guide.For the following dissolution schemes, clear stock solution should be prepared according to in vitro experiments, and then cosolvent should be added in turn:

in order to ensure the reliability of the experimental results, the clarified stock solution can be properly preserved according to the storage conditions; The working fluid for in vivo experiment is recommended to be prepared now and used on the same day;

The percentage shown in front of the following solvent refers to the volume ratio of the solvent in the final solution; If precipitation or precipitation occurs in the preparation process, it can be assisted by heating and/or ultrasound.
