Biological Activity

Description
BQ-123, a strong and specific endothelin A (ETA) receptor blocker, boasts an IC50 of 7.3 nM and a Ki of 25 nM. It hinders endothelin-1-induced growth of human pulmonary artery smooth muscle cells and reduces blood pressure in various rat hypertension models[1][2][3].
Target
  • ET-A

    Endothelin A receptor, IC50:7.3 nM

human isolated coronary artery rings 0.1 μM and 1 μM 30 min To investigate the antagonistic effects of BQ-123 on sarafotoxin S6b- and ET-1-induced contractions. Results showed that 0.1 μM BQ-123 had a stronger antagonistic effect on sarafotoxin S6b-induced contractions (pKB 7.86), while 1 μM BQ-123 had a weaker effect on ET-1-induced contractions (pKB 5.75). Animal Model
Administration Dosage Frequency Description References
Rats Anaesthetized rats Intravenous injection 1.6 µmol/kg Single injection BQ-123 alone induced a decrease in femoral mean arterial pressure accompanied by systemic vasodilation, peaking at 3 min and then slowly returning to baseline. The 1.6 μmol/kg dose significantly reduced the sustained vasoconstrictor effects of ET-1 and enhanced the early vasodilator effects of ET-1. Br J Pharmacol. 1992 Dec;107(4):912-8
Rat Anesthetized rat Intravenous injection 1 mg/kg Single dose BQ-123 (1 mg/kg, i.v.) potently inhibits the pressor response induced by ET-1 but does not affect the transient depressor response. Proc Natl Acad Sci U S A. 1994 May 24;91(11):4892-6
Mice Jmjd3fl/flMyh11CreERT mice Intraperitoneal 200 nmol/kg/day Once daily for 7 days BQ-123 reversed hypertension induced by JMJD3 deletion J Clin Invest. 2025 Mar 27;135(11):e186146
