Biological Activity

Description
DTHIB is a direct and selective inhibitor of heat shock factor 1 (HSF1) with a Kd of 160 nM for binding to the HSF1 DNA binding domain (DBD). It inhibits the HSF1 cancer gene signature (HSF1 CaSig) and selectively promotes the degradation of nuclear HSF1, exhibiting potent anticancer activities that can be utilized in prostate cancer research [1].

Concentration Treated Time Description References
22Rv1 cells 5 μM 72 hours DTHIB inhibited AR signaling and cell growth Sci Transl Med. 2020 Dec 16;12(574):eabb5647
BT-549 0 to 200 µM 72 hours To assess the effect of DTHIB on BT-549 cell viability, IC50 was determined to be 7.7 µM. Cancers (Basel). 2023 Oct 12;15(20):4965
C4-2 cells 1 μM 48 hours DTHIB inhibited the HSF1 cancer gene signature and prostate cancer cell proliferation Sci Transl Med. 2020 Dec 16;12(574):eabb5647
OVCAR-3 cells 3 μM 72 hours DTHIB pretreatment of OVCAR-3 cells prevented the rescue of miR-145-5p expression achieved by CDDP at 42 °C, and under these CDDP/DTHIB co-treatment conditions, the decrease in tumor cell proliferation was no longer observed. Cell Death Dis. 2023 Aug 19;14(8):535
BT-549 2.5 µM 7-21 days At 2.5 µM, DTHIB did not affect growth in 2D culture but formed round, tight spheroids in Matrigel, reducing the formation of stellate spheroids. Cancers (Basel). 2023 Oct 12;15(20):4965
BT-549 5 µM 48 hours DTHIB treatment led to a decrease in HSF1 levels and approximately 20% inhibition of heat-shock-induced HSPA1 expression. Cancers (Basel). 2023 Oct 12;15(20):4965

Dissolving Methods
Please choose the appropriate dissolution scheme according to your animal administration guide.For the following dissolution schemes, clear stock solution should be prepared according to in vitro experiments, and then cosolvent should be added in turn:

in order to ensure the reliability of the experimental results, the clarified stock solution can be properly preserved according to the storage conditions; The working fluid for in vivo experiment is recommended to be prepared now and used on the same day;

The percentage shown in front of the following solvent refers to the volume ratio of the solvent in the final solution; If precipitation or precipitation occurs in the preparation process, it can be assisted by heating and/or ultrasound.
