Recombinant Human ICAM-1 (C-6His, HEK293) is a human-derived protein expressed in HEK293 human cells and carries a C-terminal 6×His tag. This product corresponds to intercellular adhesion molecule 1, which is normally expressed at low levels on the membranes of leukocytes and endothelial cells. It is a crucial transmembrane protein mediating cell-cell adhesion and leukocyte transendothelial migration. The protein features highly glycosylated structural characteristics and can bind to immune-related ligands such as macrophage adhesion ligand-1, LFA-1 integrin, and fibrinogen. It also serves as a key binding site for rhinovirus on host cells.

Biological Activity

Description
Inter-Cellular Adhesion Molecule 1 (ICAM1) is a type of intercellular adhesion molecule continuously present in low concentrations in the membranes of leukocytes and endothelial cells. As an endothelial and leukocyte-associated transmembrane protein, ICAM1 is well known for its importance in stabilizing cell-cell interactions and facilitating leukocyte endothelial transmigration. The presence of heavy glycosylation and other structural characteristics lend ICAM1 binding sites for a number of immune-associated ligands. Notably, ICAM-1 binds to macrophage adhesion ligand-1 (Mac-1; ITGB2 / ITGAM), leukocyte function associated antigen-1 (LFA-1/integrin), and fibrinogen.ICAM-1 expressed by respiratory epithelial cells is also the binding site for rhinovirus, the causative agent of most common colds.
Protein Species Human
Expression System Human Cells
Tag C-6His
Protein Expression Interval Gln28-Glu480
Protein ID P05362
Endotoxin < 1 EU/µg as determined by LAL test.
Characteristics Lyophilized powder
Formulation Description Lyophilized from a 0.2 μm filtered solution of 20mM PB, 150mM NaCl, pH 7.2.
Reconstitution Method Always centrifuge tubes before opening.Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100μg/ml. Dissolve the lyophilized protein in distilled water. Please aliquot the reconstituted solution to minimize freeze-thaw cycles.
For long-term storage, it is recommended to further dilute the product with a solution containing carrier proteins (e.g., 0.1% BSA, 10% FBS, or 5% HSA). For serum-free cultures or in vivo animal experiments where BSA, FBS, or HSA cannot be present in the cytokine preparation, trehalose may be used as an alternative carrier. Aliquots should be stored frozen at -20°C to -80°C.
Note: Low-adsorption tubes are recommended for aliquot storage, and repeated freeze-thaw cycles should be avoided. The dilution concentration for aliquoting should not be lower than 10 μg/mL, and the aliquot volume should be no less than 10 μL.
Aliquoted protein remains stable for up to 3 months when stored at ≤ -20°C.
