Recombinant Human CST5 (C-6His, HEK293) is a human-derived Cystatin-D recombinant protein, expressed in HEK293 cells and tagged with a C-terminal 6×His tag. The encoding gene is located within the cystatin gene locus on human chromosome 20. This protein is predominantly found in saliva and tears, belongs to the type II cysteine protease inhibitor family, and exhibits potential inhibitory activity against oral proteases, suggesting a protective role for oral tissues.

Biological Activity

Description
Cystatin-D is a protein that in humans is encoded by the CST5 gene. The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and the kininogens. The type 2 cystatin proteins are a class of cysteine proteinase inhibitors found in a variety of human fluids and secretions. The cystatin locus on chromosome 20 contains the majority of the type 2 cystatin genes and pseudogenes. This gene is located in the cystatin locus and encodes a protein found in saliva and tears. The encoded protein may play a protective role against proteinases present in the oral cavity.
Protein Species Human
Expression System Human Cells
Tag C-6His
Protein Expression Interval Gly21-Val142
Protein ID P28325
Endotoxin < 1 EU/µg as determined by LAL test.
Characteristics Lyophilized powder
Formulation Description Lyophilized from a 0.2 μm filtered solution of 20mM Tris-HCl, 6% Trehalose, 2% Mannitol, 0.05% Tween 80, pH8.5.
Reconstitution Method Always centrifuge tubes before opening.Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100μg/ml. Dissolve the lyophilized protein in distilled water. Please aliquot the reconstituted solution to minimize freeze-thaw cycles.
For long-term storage, it is recommended to further dilute the product with a solution containing carrier proteins (e.g., 0.1% BSA, 10% FBS, or 5% HSA). For serum-free cultures or in vivo animal experiments where BSA, FBS, or HSA cannot be present in the cytokine preparation, trehalose may be used as an alternative carrier. Aliquots should be stored frozen at -20°C to -80°C.
Note: Low-adsorption tubes are recommended for aliquot storage, and repeated freeze-thaw cycles should be avoided. The dilution concentration for aliquoting should not be lower than 10 μg/mL, and the aliquot volume should be no less than 10 μL.
Aliquoted protein remains stable for up to 3 months when stored at ≤ -20°C.
