Recombinant Human CD32b (C-6His, HEK293) is a human-derived CD32b protein expressed in HEK293 cells with a C-terminal 6×His tag. This protein corresponds to FcγRIIB, a low-affinity IgG Fc receptor featuring an immunoreceptor tyrosine-based inhibitory motif (ITIM). Upon ligand binding, it transduces inhibitory signals and plays a critical role in modulating B cell antibody production and dendritic cell activation. As a potential target for tumor-targeted therapy, it exerts significant immune-suppressive regulatory functions.

Biological Activity

Description
FcγRIIB is a low affinity receptor that recognizes the Fc portion of IgG. The human CD32 group consists of FcγRIIA, FcγRIIB, and FcγRIIC proteins that share 94-99% sequence identity in their extracellular domains but differ substantially in their transmembrane and cytoplasmic domains. FcγRII protein is expressed on cells of both myeloid and lymphoid lineages as well as on cells of non-hematopoietic origin. FcγRIIB has an intrinsic cytoplasmic immunoreceptor tyrosine-based inhibitory motif (ITIM) and delivers an inhibitory signal upon ligand binding. Ligation of FcγRIIB on B cells down-regulates antibody production and in some circumstances may promote apoptosis. Co-ligation of FcγRIIB on dendritic cells inhibits maturation and blocks cell activation. FcγRIIB may also be a target for monoclonal antibody therapy for malignancies. FcγRIIB plays an important negative-regulating role through modulating the signals from activation receptors.

Expression System Human Cells
Tag C-6His
Protein Expression Interval Thr43-Pro217
Protein ID P31994
Endotoxin < 1 EU/µg as determined by LAL test.
Characteristics Lyophilized powder
Formulation Description Lyophilized from a 0.2 μm filtered solution of 20mM PB, 150mM NaCl, pH 7.2.
Reconstitution Method Always centrifuge tubes before opening.Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100μg/ml. Dissolve the lyophilized protein in distilled water. Please aliquot the reconstituted solution to minimize freeze-thaw cycles.
For long-term storage, it is recommended to further dilute the product with a solution containing carrier proteins (e.g., 0.1% BSA, 10% FBS, or 5% HSA). For serum-free cultures or in vivo animal experiments where BSA, FBS, or HSA cannot be present in the cytokine preparation, trehalose may be used as an alternative carrier. Aliquots should be stored frozen at -20°C to -80°C.
Note: Low-adsorption tubes are recommended for aliquot storage, and repeated freeze-thaw cycles should be avoided. The dilution concentration for aliquoting should not be lower than 10 μg/mL, and the aliquot volume should be no less than 10 μL.
Aliquoted protein remains stable for up to 3 months when stored at ≤ -20°C.
