Recombinant Human KNG1 (C-6His, HEK293) is a secreted human kallikrein-binding protein expressed in HEK293 cells with a C-terminal 6×His tag. It contains three cysteine protease inhibitor domains and exists in both high-molecular-weight (HMW) and low-molecular-weight (LMW) splice isoforms. This protein is involved in coagulation and inflammatory responses, regulates the localization of prekallikrein and clotting factors, and inhibits platelet aggregation. Its bioactive peptide, bradykinin, exerts multiple physiological effects, including modulation of vascular permeability, blood pressure, and inflammation, and possesses cardioprotective properties.

Biological Activity

Description
Kininogen-1 is a secreted protein which contains three cystatin domains. There are two alternatively spliced forms, designated as the high molecular weight (HMW) and low MW (LMW) forms. Kininogen-1 plays a critical role in blood coagulation and inflammatory response. Kininogens are inhibitors of thiol proteases. Kininogen-1 participates in blood coagulation by helping to position optimally prekallikrein and factor XI next to factor XII, also inhibits the thrombin- and plasmin-induced aggregation of thrombocytes. The active peptide bradykinin that is released from Kininogen-1 shows a variety of physiological effects: influence in smooth muscle contraction, induction of hypotension, natriuresis and diuresis, decrease in blood glucose level. It is a mediator of inflammation and causes increase in vascular permeability, stimulation of nociceptors release of other mediators of inflammation. It has a cardioprotective effect. LMW-kininogen inhibits the aggregation of thrombocytes and doesn’t involved in blood clotting.
Protein Species Human
Expression System Human Cells
Tag C-6His
Protein Expression Interval Gln19-Ser427
Protein ID P01042-2
Endotoxin < 1 EU/µg as determined by LAL test.
Characteristics Lyophilized powder
Formulation Description Lyophilized from a 0.2 μm filtered solution of 20mM Hac-NaAC, 150mM NaCl, pH 4.0.
Reconstitution Method Always centrifuge tubes before opening.Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100μg/ml. Dissolve the lyophilized protein in distilled water. Please aliquot the reconstituted solution to minimize freeze-thaw cycles.
For long-term storage, it is recommended to further dilute the product with a solution containing carrier proteins (e.g., 0.1% BSA, 10% FBS, or 5% HSA). For serum-free cultures or in vivo animal experiments where BSA, FBS, or HSA cannot be present in the cytokine preparation, trehalose may be used as an alternative carrier. Aliquots should be stored frozen at -20°C to -80°C.
Note: Low-adsorption tubes are recommended for aliquot storage, and repeated freeze-thaw cycles should be avoided. The dilution concentration for aliquoting should not be lower than 10 μg/mL, and the aliquot volume should be no less than 10 μL.
Aliquoted protein remains stable for up to 3 months when stored at ≤ -20°C.
