Recombinant Human DKK-1 (N-8His, HEK293) is a human-derived Dickkopf-related protein 1 (DKK-1) expressed in HEK293 cells and fused with an N-terminal 8×His tag. As a member of the Dickkopf family, it antagonizes the canonical Wnt signaling pathway by inhibiting the binding of LRP5/6 to Wnt and forming complexes with KREMEN. This protein plays critical roles in embryonic development, bone metabolism, tumorigenesis, and Alzheimer's disease.

Biological Activity

Description
Dickkopf-related protein 1(DKK-1), is a member of the dickkopf family. DKK1 secreted proteins with two cysteine-rich domains separated by a linker region. It antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease.

Formulation Description Lyophilized from a 0.2 μm filtered solution of PBS, pH 7.4.
Reconstitution Method Always centrifuge tubes before opening.Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100μg/ml. Dissolve the lyophilized protein in distilled water. Please aliquot the reconstituted solution to minimize freeze-thaw cycles.
For long-term storage, it is recommended to further dilute the product with a solution containing carrier proteins (e.g., 0.1% BSA, 10% FBS, or 5% HSA). For serum-free cultures or in vivo animal experiments where BSA, FBS, or HSA cannot be present in the cytokine preparation, trehalose may be used as an alternative carrier. Aliquots should be stored frozen at -20°C to -80°C.
Note: Low-adsorption tubes are recommended for aliquot storage, and repeated freeze-thaw cycles should be avoided. The dilution concentration for aliquoting should not be lower than 10 μg/mL, and the aliquot volume should be no less than 10 μL.
Aliquoted protein remains stable for up to 3 months when stored at ≤ -20°C.
