Recombinant Mouse DNAM-1 (C-6His, HEK293) is a mouse-derived recombinant protein expressed in HEK293 human cells and tagged with a C-terminal 6×His tag. This type I transmembrane glycoprotein belongs to the immunoglobulin superfamily and functions as an activating receptor. It interacts with ligands CD155 and CD112 to activate natural killer (NK) cells, promote immune cell activation and dendritic cell maturation, facilitate platelet adhesion, and inhibit osteoclast formation. The protein is involved in tumor metastasis processes.

Synonyms: Recombinant Mouse DNAM-1 (C-6His, HEK293); CD226 antigen; platelet and T cell activation antigen 1

Biological Activity

Description
Mouse DNAX accessory molecule-1(DNAM-1) is a type I transmembrane glycoprotein that belongs to the immunoglobulin superfamily. As an activating receptor, it interacts with the ligands CD155 and CD112, and activates natural killer (NK) cells via its immu-noreceptor tyrosine-based activatory motif (ITAM). Mature mouse DNAM-1 has extracellular domain (ECD) that contains two Ig-like C2-set domains, and possesses a cytoplasmic region that contains motifs for binding PDZ domains. DNAM-1 is expressed on several lymphoid and myeloid cell types and interacts with CD155/PVR and Nectin-2/CD112. Ligation of DNAM-1 promotes the activation of NK cells, CD8+ T cells, and mast cells, induces dendritic cell maturation, initiates megakaryocyte and activated platelet adhesion to vascular endothelial cells, and stimulates monocyte extravasation; Conversely, it inhibits the formation of osteoclasts. Platelet-endothelium interactions that are mediated by DNAM-1 enable the metastasis of tumor cells to the lung.
Protein Species Mouse
Expression System Human Cells
Tag C-6His
Protein Expression Interval Glu19-Pro254
Protein ID Q8K4F0
Endotoxin < 1 EU/µg as determined by LAL test.
Characteristics Lyophilized powder
Formulation Description Lyophilized from a 0.2 μm filtered solution of PBS, pH 7.4.
Reconstitution Method Always centrifuge tubes before opening.Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100μg/ml. Dissolve the lyophilized protein in distilled water. Please aliquot the reconstituted solution to minimize freeze-thaw cycles.
For long-term storage, it is recommended to further dilute the product with a solution containing carrier proteins (e.g., 0.1% BSA, 10% FBS, or 5% HSA). For serum-free cultures or in vivo animal experiments where BSA, FBS, or HSA cannot be present in the cytokine preparation, trehalose may be used as an alternative carrier. Aliquots should be stored frozen at -20°C to -80°C.
Note: Low-adsorption tubes are recommended for aliquot storage, and repeated freeze-thaw cycles should be avoided. The dilution concentration for aliquoting should not be lower than 10 μg/mL, and the aliquot volume should be no less than 10 μL.
Aliquoted protein remains stable for up to 3 months when stored at ≤ -20°C.
