Recombinant Human DNAM-1 (C-Fc, HEK293) is a human-derived DNAM-1 protein expressed in HEK293 cells with a C-Fc tag. This 65 kDa type I transmembrane glycoprotein belongs to the immunoglobulin superfamily. Its mature form contains an extracellular region with two Ig-like C2-set domains and an intracellular region featuring binding motifs for PDZ and band 4.1 family proteins. It is broadly expressed on lymphoid and myeloid cells, interacts with CD155 and CD112, and promotes activation of NK cells and CD8+ T cells. The protein is involved in immune responses, vascular adhesion, and tumor metastasis.

Description
Human DNAX accessory molecule 1 (DNAM-1/CD226) is a 65 kDa type I transmembrane glycoprotein in the immunoglobulin superfamily. Mature human DNAM-1 contains an extracellular domain (ECD) with two Ig-like C2-set domains and a cytoplasmic region that contains motifs for binding PDZ domains and band 4.1 family proteins. DNAM-1 is expressed on multiple lymphoid and myeloid cells and interacts with CD155 and CD112. Ligation of DNAM-1 promotes the activation of NK cells, CD8+ T cells, and mast cells, dendritic cell maturation, megakaryocyte and activated platelet adhesion to vascular endothelial cells, and monocyte extravasation; it inhibits the forrmation of osteoclasts. Plateletendothelium, interactions mediated by DNAM-1, enable the metastasis of tumor cells to the lung.

Formulation Description Lyophilized from a 0.2 μm filtered solution of PBS, pH 7.4.
Reconstitution Method Always centrifuge tubes before opening.Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100μg/ml. Dissolve the lyophilized protein in distilled water. Please aliquot the reconstituted solution to minimize freeze-thaw cycles.
For long-term storage, it is recommended to further dilute the product with a solution containing carrier proteins (e.g., 0.1% BSA, 10% FBS, or 5% HSA). For serum-free cultures or in vivo animal experiments where BSA, FBS, or HSA cannot be present in the cytokine preparation, trehalose may be used as an alternative carrier. Aliquots should be stored frozen at -20°C to -80°C.
Note: Low-adsorption tubes are recommended for aliquot storage, and repeated freeze-thaw cycles should be avoided. The dilution concentration for aliquoting should not be lower than 10 μg/mL, and the aliquot volume should be no less than 10 μL.
Aliquoted protein remains stable for up to 3 months when stored at ≤ -20°C.
