Pactimibe sulfate (CS-505) is a dual ACAT1/2 inhibitor with IC50 values of 4.9 μM for ACAT1 and 3.0 μM for ACAT2. It has IC50 values of 2.0 μM in hepatocytes, 2.7 μM in macrophages, and 4.7 μM in THP-1 cells. Pactimibe sulfate non-competitively inhibits oleoyl-CoA with a Ki of 5.6 μM and significantly inhibits cholesterol ester formation with an IC50 of 6.7 μM. It has the potential for anti-atherosclerosis.
