94% With dmap; 1-ethyl-(3-(3-dimethylamino)propyl)-carbodiimide hydrochloride; In dichloromethane; at 20℃; General procedure: EDCI (2.0 eq) was added to a solution of 1-benzyl-3-(tert-butyl)-1H-pyrazole-5-carboxylicacid (1.0 eq) and DMAP (2.0 eq) in dichloromethane. The reaction mixture was stirred for 10- 15 minutes, after which the appropriate amino cyano analogue (1.2 eq) was added. The resulting reaction mixture was stirred for 4 - 20 h before being washed with saturated aqueous solutions of NaHCO3 (3 ×), NH4Cl (2 ×), and NaCl (2 ×). The organic layer was dried (MgSO4), filtered and concentrated in vacuo. The crude mixture was chromatographed (SiO2, EtOAc-hexane, 10:90) to give the desired nitrile compound.
