A solution of <strong>[1192711-71-9]2,4-dichloro-6-methyl-7H-pyrrolo[2,3-d]pyrimidine</strong> (190 mg, 0.94 mmol), cyclobutylamine (0.160 mL, 1.88 mmol) and triethylamine (0.200 mL, 1.44 mmol) in nBuOH (6 mL) was stirred at 70 0C overnight. It was then purified by HPLC to give 2- chloro-N-cyclobutyl-6-methyl-7H-pyrrolo[2,3-d]pyrimidin-4-amine (100 mg). |

A mixture of 6-methyl-7H-pyrrolo[2,3-d]pyrimidine-2,4-diol (490 mg, 2.97 mmol) in phenylphosphonic dichloride (10 mL) was heated at 180 0C for 5 h. It was poured onto ice. Et20 (100 mL) was added. The organic phase was separated, washed with 5% NaHCO3, dried over Na2SO4, concentrated in vacuo to give 2,4-dichloro-6-methyl-7H-pyrrolo[2,3- d]pyrimidine as a solid (380 mg). |

General procedure: A mixture of 2,4-dichloro-7H-pyrrolo[2,3-d]pyrimidine 18a (51.3 g) and 1.36 L of 1 N NaOH was stirred at 80 C overnight. The solution was subsequently chilled and adjusted to pH 6 with AcOH. The resulting precipitate was collected, washed with water and dried to afford 19a as solid (37.3 g, yield: 80.7%). |

This compound was prepared following a similar protocol as that presented for Compound 57, except that the starting material used was <strong>[1192711-71-9]2,6-dichloro-8-methyl-7-deazapurine</strong>. Briefly, 2,6- dichloro-8-methyl-7-deazapurine (50 mg, 0.25 mmol) was suspended in 1-butanol (3 mL), and DCM (2 mL) was added dropwise until the suspension became a clear solution at room temperature. Pyrrolidine (0.55 mmol) was then added slowly, and the reaction was stirred overnight until TLC analysis (DCM: MeOH 2.5%) indicated it was complete. The mixture was concentrated and the residue dissolved in DCM (20 mL), and the resulting solution was washed with water (2 x 3 mL) to remove the hydrochloride. The solution was then dried over anhydrous Na2S04 and purified by silica gel column chromatography to afford Compound 58 (55 mg, 94% yield); LC-MS m/z = 237.1 [M+l], corresponding CnHi3ClN4. |
