55% In tetrahydrofuran; at -10 - 25℃; for 16.33h; A 10 L round- bottomed flask equipped with an overhead stirrer was charged withethynylmagnesiumbromide (3.50 L, 0.5 M in THF). The flask was purged with nitrogen and cooled to-10 C before adding 1-methylpyrrolidine-2,3-dione (120 g, 1.06 mol) over the course of 20 min. The resultant mixture was warmed to 20 - 25 C and stirred for 16 h. The resulting mixture was quenched with aq. NH4Cl (120 g in 360 mL H2O) followed by dilution with DCM (3.50 L). After being slurried for 1 h, the suspension was filtered, and the filtrate was dried over anhydrous Na2SO4 (500 g) and treated with activated charcoal (24 g). The activated charcoal was removed by filtration and the filtrate was concentrated under vacuum to dryness. The residue was slurried in MTBE (360 mL) at 20 -25 C for 1 h. The product was isolated by filtration followed by drying to give (rac)-3-ethynyl-3-hydroxy-1-methylpyrrolidin-2-one (81 g, 55%) as a yellow solid. MS (ESI): mass calcd. for C7H9NO2, 139.1; m/z found, 140.1 [M+H]+.1H NMR (300 MHz, CD3OD) d 3.40 (dd, J = 7.7, 5.3 Hz, 2H), 3.03 (s, 1H), 2.88 (s, 3H), 2.52-2.41 (m, 1H), 2.21 (dt, J = 12.7, 7.7 Hz, 1H).
