74% With dichloro(1,1'-bis(diphenylphosphanyl)ferrocene)palladium(II)*CH2Cl2; sodium carbonate; In water; 1,2-dichloro-ethane; at 120℃; for 0.25h;Microwave irradiation; To a mixture of tert-butyl 4-(5-bromo-3-cyanopyridin-2-yl)piperazine- 1-carboxylate (1.0 equiv.), methyl 4-methyl-3-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2- yl)benzoate (1.1 equiv.) and PdCl2(dppf).CH2Cl2 adduct (0.08 equiv.) in DME (0.36 M) was added Na2CO3 (3.0 equiv, 2M aqueous solution). The mixture was stirred at 120 C in the microwave for 15 min. LC-MS showed complete conversion. Added brine and EtOAc, the organic layer was dried over sodium sulfate and concentrated and purified by ISCO (0-100% EtOAc/Heptane) to give tert-butyl 4-(3-cyano-5-(5-(methoxycarbonyl)-2- methylphenyl)pyridin-2-yl)piperazine-1-carboxylate in 74% yield. LCMS (m/z) (M+H) = 381.2, Rt = 1.13 min.
