YieldReaction ConditionsOperation in experiment
67% To a stirred solution of dimethyl malonate (0.39 mL, 3.429 mmol, CAS RN 108-59-8) in THF (5 mL) was added sodium hydride (60% in mineral oil) (171.4 mg, 4.286 mmol, CAS RN 7646-69- 7) at 0C. The reaction mixture was stirred at 25C for 30 mm., then cooled again to 0C and 1- chloro-2-fluoro-3-nitrobenzene (500 mg, 2.86 mmol, CAS RN 2106-49-2) was added. Thereaction mixture was stirred at 25 C for 16 h. After completion of the reaction, it was quenchedwith saturated aqueous NH4C1 solution (10 mL) and was extracted with EtOAc (2x50 mL). The organic layer washed with brine, dried over Na2504 and concentrated under reduced pressure.The crude product was purified using silica gel column chromatography (10-12% EtOAc/nhexane) to get the title compound as an off-white solid (550 mg, 67%).
