YieldReaction ConditionsOperation in experiment
3.9 g to 2-(S)-aminomethyl-pyrrolidine-l-carboxylic acid benzyl ester (4.68g, 20 mmol) in DCM (30 mL), formic acid cyanomethyl ester (2.0g, 23.5 mmol) was added drop-wise. The mixture was then stirred at rt for 1 day, concentrated and re-dissolved in DCM (60 ml). To this solution, Burgess reagent (4.7g, 19.7 mmol) was added. The mixture was heated to reflux for lh. Additional Burgess reagent (2.38 g, 10 mmol) was added and it was refluxed for another lh. The mixture was then concentrated and purified by silica gel chromatography eluted with DCM/EtOAc to afford the desired product as yellowish oil (3.9 g). MS (M+l)+: 245.3.
