YieldReaction ConditionsOperation in experiment
With N-ethyl-N,N-diisopropylamine; HATU; In N,N-dimethyl-formamide; at -20 - 20℃; for 1h; Step 1 : HATU (3.56 g, 9.35 mmol, 1.15 eq.) was added to a solution of amine 14-1 (1.8 g, 8.13 mmol, 1.0 eq.), 1-cyanocyclopropanecarboxylic acid (0.90 g,8.13 mmol, 1.0 eq.) and N-ethyl-N-isopropylpropan-2-amine (DIPEA; 3.15 g, 8.13 mmol, 1.0 eq.) in DMF (80 mL) at -20°C. The reaction mixture was stirred at room temperature for one hour. Ethyl acetate was added, the organic phase was washed with a saturated NaHC03 solution, dried with anhydrous MgS04 and concentrated under reduced pressure to give 1.83 g (72percent) of crude Intermediate 14-2. This was used as such in the next step.
