97% With hydrogen;10Mol% palladium on carbon; In ethanol; dichloromethane; for 0.5h; Preparation of 4-bromo-5-(trifluoromethyl)benzene-1,2-diamine (18).; 10% Pd/C (373 mg, 0.4 mmol, 10 mol % Pd) was added to compound 17 (1.0 g, 3.5 mmol) in ethanol:dichloromethane (8:1, 140 mL). The mixture was degassed and placed under H2 (1 atm) and stirred for 30 min. The mixture was filtered through a 0.45 mum syringe filter and concentrated under reduced pressure to provide 18 (865 mg, 97%) as an oil: LRESIMS m/z 255.0 [M+H]+, calcd. for C7H7Br1F3N2 255.0.
With ammonium chloride; zinc; In ethanol; at 20℃; for 16.0h; Compound 11b (2.85 g, 0.01 mol) was dissolved in 30 mL of ethanol. Zinc powder (5.9 g, 0.09 mol) was added in portions followed by the addition of ammonium chloride (1.07 g, 0.02 mol, 2 eq). The reaction mixture was stirred at room temperature overnight (16 h). The reaction mixture was filtered over a pad of celite and the solvent was evaporated in vacuo. The residue was taken up in ethyl acetate (40 mL), washed with 35 mL of brine, dried over Na2SO4, and evaporated in vacuo. The residue was purified by chromatography (silica gel, CH2Cl2:MeOH, 96:4-94:6)) to provide the title Compound 11c as a bright yellow solid (1.53 g), which was used without further purification in the next step. 1H-NMR (400 MHz, CD3OD) delta (ppm): 6.99 (s,1H) 6.92 (s,1 H) MS (ESI, pos. ion) m/z: 254.7 (M+1).
With tin(II) chloride hydrate; In ethanol; ethyl acetate; at 75 - 80℃; for 4.0h; To the solution of <strong>[683241-86-3]4-bromo-2-nitro-5-(trifluoromethyl)aniline</strong> (3.19 g, 11.2 mmol) in 35 ml of ethyl acetate and 17.5 ml of ethanol was added tin(II) chloride hydrate. The mixture was heated at 75-80 C. for 4 h, cooled, poured onto crushed ice (200 g) and neutralized with saturated sodium bicarbonate aqueous solution to pH8-9. The solid was removed by filtration thorough a Celite pad and washed with ethyl acetate (150 ml). The organic was separated from the filtrate and the aqueous phase was extracted with ethyl acetate twice. The organic was combined, washed with brine, dried over magnesium sulfate and concentrated in vacuo to afford the title product as a brown crystalline solid. LC-MS: 280.87(M+), 2.02 min (LC/MS method A)
