YieldReaction ConditionsOperation in experiment
With potassium permanganate; water; for 19h;Reflux; Preparation 84-Chloro-3-methoxy-pyridine-2-carboxylic acid; A suspension of <strong>[107512-34-5]4-chloro-3-methoxy-2-methyl-pyridine</strong> (prepared as described in Journal of Medicinal Chemistry, 1992, pages 1049-1057; 8g, 51 mmol) in water (30OmL) was treated with potassium permanganate (9.6g, 61 mmol) and heated at reflux. Two more batches of potassium permanganate (2 x 9.6g, 2 x 61 mol) were added to the reaction mixture at 1.5 hour intervals and the reaction mixture was then refluxed for another 16 hours. The reaction mixture was filtered through a short bed of Celite and the filtrate was extracted with dichloromethane (3 x 5OmL). The aqueous layer was acidified with citric acid and then extracted with a mix of methanol:dichloromethane (1 :9) (1 O x 10OmL). Combined organic phases were washed with saturated aqueous sodium chloride solution (1 x 20OmL), dried (sodium sulfate), and then evapourated in vacuo to furnish the product as a white solid, 4.6g.1 H NMR (400MHz, DMSOd6): = 3.88 (3H, s), 7.75 (1 H, d), 8.33 (1 H, d), 13.70 (1 H, bs) ppm. LRMS: 188 [M+H]+.
