28.5% In isopropyl alcohol; at 90℃; for 16h; Tert-butyl N-(2-amino-2-thioxo-ethyl)carbamate (489.4 mg, 2.57 mmol) was added to a stirred solution of benzyl 3-bromo-4-oxo-piperidine-1-carboxylate (800 mg, 2.57 mmol) in IPA (10 mL) at RT. The reaction mixture was stirred at 90C for 16 h and concentrated under reduced pressure. The residue was diluted with water (5 mL), acidified (pH--2) with iN HC1 and extracted with Et20 (2x20 mL). The aqueous phase was basified (pH-8) with saturatedNaHCO3 solution and extracted with DCM (2x50 mL). The combined organic layer was washed with brine, dried over Na2SO4 and concentrated under reduced pressure affording a pale brown solid (220 mg, 28.5%). M/z 304.3 (M+H).
