YieldReaction ConditionsOperation in experiment
94% With lithium aluminium tetrahydride; In tetrahydrofuran; at 0℃; for 1h; The methyl ester intermediate 42 (2.09 g, 10.29 mmol) was added slowly to a suspension of LiAlH4 (1.18 g, 31.09 mmol) in tetrahydrofuran (20 mL) at 0 C. The reaction mixture was stirred at 0 C for 1 h. The reaction was quenched by adding H2O (1.5 g) and CH3OH (20 mL) slowly, then filtered, concentrated to afford the residue. DCM (20 mL) and H2O (20 mL) were added, the organic phase was separated and aqueous layer was extracted with DCM (20 mL × 2). The combined organic layers were dried over Na2SO4. The solvent was concentrated to afford primary alcohol intermediate 43 (1.7 g, 94%) as a light yellow solid.
