YieldReaction ConditionsOperation in experiment
58% With sodium carbonate; In 1,4-dioxane; water; at 50℃; for 20h; A solution of succinimidyl 2,2-diphenylacetate (2) (5.83 g, 18.80 mmol, 1 equiv.) in dioxane (80 mL) was added drop-wise to a solution of (R)-Ndelta-benzyloxycarbonylornithine (1) (5 g, 18.80 mmol, 1 equiv.), and Na2CO3 (2 g, 18.80 mmol, 1 equiv.)in a mixture of water and 1,4-dioxane (2.5/1, v/v, 100 mL) over a period of 30 min and the mixture was stirred at 50 C for 20 h. The resultant slurry was filtered to remove the remaining starting materials and the filtrate evaporated to dryness. Ethylacetate (EtOAc) (50 mL) was added and washed with 1M HCl and brine (210 mL, each) and the aqueous washes were re-extracted with a new portion of EtOAc (30 mL). The organic layers were combined, dried over anhydrous Na2SO4, filtered, and evaporated under reduced pressure. Purification with silica gel column chromatography (eluent: 50-100 % EtOAc/n-hexane) afforded the product as a light yellow oil (2.52 g, 5.48 mmol, 58 %). deltaH (d6-DMSO, 400.13 MHz) 1.36-1.1.47 (m, 2H), 1.51-1.72 (m, 2H), 2.98 (m, 2H), 4.19 (m, 1H), 5.01 (s, 2H), 5.10 (s, 1H), 7.17-7.39 (m, 16H), 8.52 (AB doublet, 1H), 12.57 (br s, 1H). deltaC (CDCl3, 100.61 MHz) 173.90, 171.58, 156.56, 140.84, 140.76, 137.73, 129.02, 128.98, 128.81,128.66, 128.59, 128.21, 128.16, 127.07, 127.01. m/z 482.99 ([M Na]), 943.29 ([2M Na]).
