98% With potassium hydrogencarbonate; In tetrahydrofuran; water; at 20℃; for 14h; A solution of Cbz-Phe-OSu (1.00 g, 2.52 mmol) and HCys(Trt)-OH (963 mg, 2.65 mmol) in 1 M aq. KHCO3:THF (1:1, 12.6 mL) was stirred vigorously for 14 h overnight. Upon completion of the reaction, the mixture was acidified to pH 3 with 10percent citric acid (30 mL) and the aqueous layer extracted with EtOAc (3 x 60 mL). The combined organic extracts were washed with brine, dried with Na2SO4, and concentrated in vacuo. Purification of the resulting white foam by silica gel chromatography (10percent MeOH/CHCl3) afforded the dipeptide acid (3.20 g, 4.96 mmol, 98percent) as a white foam.
