With tetrakis(triphenylphosphine) palladium(0); potassium carbonate; In 1,2-dimethoxyethane; water; at 90℃;Inert atmosphere; A mixture of (6-chl oropyri din-3 -yl)m ethanol 49A (0.75 g, 5.26 mmol), 4-(trifluoromethyl)phenylboronic acid (1 g, 5.26 mmol), K2CO3 (2.18 g, 15.78 mmol), and Pd(PPh3)4 (607 mg, 0.526 mmol) in DME-H2O (24 mL, 5: 1 in volume) was heated at 90 C under nitrogen overnight. The mixture was cooled down to room temperature, diluted with ethyl acetate (50 mL), washed with water (50 mL x 2) and brine (50 mL), dried over anhydrous sodium sulfate, filtered, and concentrated to give a crude product, which was purified with column chromatography on silica gel (ethyl acetate in petroleum ether, from 0% to 50% v/v) to yield Compound 49B: LC-MS (ESI) m/z: 254 [M+H]+.
