YieldReaction ConditionsOperation in experiment
55% 8-Methoxy quinoline carboxaldehyde 1 was synthesized as reported.1 Compound 1 was subjected to Wittig-Horner reaction with diethyl 2-nitrobenzylphosphonate as follows. In a 50 ml round bottom flask, diethyl 2-nitrobenzyl phosphonate (263 mg, 0.96 mmol) and NaH (52 mg, 1.2 mmol) were stirred in 20 ml dry THF under argon atmosphere for 30 min. Compound 1 (150 mg, 0.81 mmol) dissolved in 15 ml dry THF was added to the above mixture drop wise in one hour and stirring was continued for 6 h at room temperature. After evaporation of the solvent under vacuum, water (30 ml) was added and the reaction mixture was extracted three times each with 20 ml chloroform. The organic layers were combined and washed once with saturated NaCl solution and then dried over anhydrous Na2SO4 and column chromatography on silica gel (60-120 mesh) in 30% ethyl acetate-hexane yielded the product as yellow solid (137 mg, 55%). mp. 92C. IR (KBr): max 2970, 1603, 1560, 1514, 1462, 1429, 1377, 1346, 1332, 1261, 1109, 993, 962, 825, 754 cm-1. 1H NMR (500 MHz, CD3CN): (ppm) 8.24 (s, 1H, J = 8.5 Hz), 8.06 (d, 1H, J = 16 Hz), 8.02-7.99 (m, 2H), 7.79 (d, 1H, J = 8.5 Hz), 7.75-7.73 (m, 1H), 7.57-7.53 (m, 1H), 7.51-7.45 (m, 2H), 7.45 (d, 1H, J = 16 Hz) 7.17 (dd, J1,2 = 1.5 Hz, J1,3 = 7.5 Hz, 1H),4.03 (s, 3H, CH3).13C NMR (125 MHz, CD3CN) : 156.90, 155.09,137.95, 135.05, 134.84, 133.19, 130.50, 139.10, 130.03, 129.76, 128.41, 126.02, 121.52, 120.75, 118.69, 110.03, 56.88. MS (FAB) m/z calcd. 306.32, found 306.36.
