With acetic acid; at 110℃; for 2h; [00548] To a mixture of ethyl 2-amino-lH-pyrrole-3-carboxylate (308 mg, 2.0 mmol) in AcOH (10 mL) at 110 C was added the solution of 3-oxobutanal in EtOAc. The reaction mixture was stirred at 110 C for 2 h, then cooled to RT and concentrated in vacuo. The resulting residue was purified by silica gel column (EA:PE; 1 : 1) to afford ethyl 4- methylpyrrolo[l,2-a]pyrimidine-8-carboxylate (57 mg, 14%) as a brown solid. 1H NMR (500 MHz, CDC13): delta 8.44 (d, J= 4.0 Hz, 1H), 7.52 (d, J= 3.0 Hz, 1H), 7.12 (d, J= 3.5 Hz, 1H), 6.66 (d, J= 4.0 Hz, 1H), 4.45 (q, J= 7.0 Hz, 2H), 2.64 (s, 3H), 1.44 (t, J= 7.0 Hz, 3H). LC-MS m/z: 205.2 [M+H]+. Purity (214 nm): > 69%; tR = 1.45 min
