General procedure: 2,4-Dichloro-5-fluoropyrimidine (2) was synthesized from5-FU by treating with phosphorus oxychloride and N,Ndimethylanilinein good yield. Compound 2 was hydrolyzedin tetrahydrofuran by adding 2 N sodium hydroxide to give3, which was treated with sodium alkoxide to form 2-butoxy-5-fluoropyrimidine-4(3H)-one (4). The spectra data ofcompounds 2-4 are in good agreement with literature.22,26,27 |

Procedure for Preparation of 2-Butoxy-4-chloro-5-fluoropyrimindine 5.
A stirred mixture of compound 4(16.0 g, 86 mmol) and phosphorus oxychloride (39.5 g, 260mmol) was heated to 95 °C and N,N-dimethylaniline (20.7 g,171 mmol) was added dropwise over 0.5 h. The mixture wasmaintained at 95 °C for 15 h before cooling to room temperature,and cautiously quenched by 3 N aqueous hydrochloric acid (45 mL), keeping the temperature below 30 °C over 4 h.The mixture was extracted with dichloromethane (80 mL),and the organic layer was washed with water (30 mL) andconcentrated under reduced pressure.
