YieldReaction ConditionsOperation in experiment
100% To a solution of 2-methylsulfanyl-8H-pyrido[2,3-d]pyrimidin- 7-one (300 mg, 1.55 mmol) in anhydrous dimethylformamide (10 mL) sodium hydride (60% in mineral oil, 93 mg, 2.33 mmol) was added and stirred at room temperature. After 30 min, a solution of 3-bromo-2-chloromethyl-thiophene (426 mg, 2.01 mmol) in anhydrousdimethylformamide (1 mL) was added slowly and the stirring was continued for 18 h. The resulting mixture was poured into ice water (50 g) then extracted with ethyl acetate (3 x 30 mL). The combined organic layers was dried over sodium sulfate, filtered and evaporated. The title compound (686 mg, 1.86 mmol, quant.) was obtained as a light brown solid. ESMS m/z 368 (M+H)+.
100% [00475] To a solution of [211244-81-4]2-methylsulfanyl-8H-pyrido[2,3-d]pyrimidin-7-one (1) (300 mg, 1.55 mmol) in anhydrous dimethylformamide (10 mL) sodium hydride (60% in mineral oil, 93 mg, 2.33 mmol) was added and stirred at room temperature. After 30 min, a solution of 3-bromo-2- chloromethyl-thiophene (8) (426 mg, 2.01 mmol) in anhydrous dimethylformamide (1 mL) was added slowly and the stirring was continued for 18 h. The resulting mixture was poured into ice water (50 g) then extracted with ethyl acetate (3 x 30 mL). The combined organic layers was dried over sodium sulfate, filtered and evaporated.
