YieldReaction ConditionsOperation in experiment
With acetic acid; for 3h;Reflux; General procedure: To a stirred solution of 35b (3.0 g, 13.8 mmol) in AcOH (20 mL) was added ethyl 2-oxocyclopentanecarboxylate (2.37 g, 15.2 mmol) at room temperature and the reaction mixture was allowed to be heated at reflux and stirred for 3 h. The reaction mixture was cooled to room temperature and diluted with Et2O/hexane (1:3). The precipitates were collected by filtration to give 3-(2-methyl-4-methoxyphenyl)-2-methyl-6,7-dihydro-5H-cyclopenta[d]pyrazolo[1,5-a]pyrimidin-8-ol (3.88 g, 91% yield) as an off-white powder, which was used for the next reaction without further purification.
