75% With potassium carbonate; In tetrahydrofuran; at 65℃; for 16h; The 4-(2-hydroxyethyl)-l-<strong>[59702-07-7]methylpiperazin-2-one</strong> used as starting material was prepared as follows :-2-Bromoethanol (5.60 mL, 78.85 mmol) was added to 1 -<strong>[59702-07-7]methylpiperazin-2-one</strong> (1.80 g, 15.77 mmol) and potassium carbonate (6.54 g, 47.31 mmol) in THF (20 mL). The resulting mixture was stirred at 65°C for 16 hours. The mixture was cooled to room temperature, fltered and the solvents evaporated to give crude product. The crude product was purified by flash silica chromatography, elution gradient 0 to 8percent MeOH in DCM. Pure fractions were evaporated to dryness to give 4-(2-hydroxyethyl)-l-<strong>[59702-07-7]methylpiperazin-2-one</strong> (1.870 g, 75.0percent). IH NMR (399.9 MHz, CDC13) delta 2.55 (2H, t), 2.71 (2H, t), 2.90 (3H, s), 3.15 (2H, s), 3.28 (2H, t), 3.60 (2H, t)
With potassium carbonate; In tetrahydrofuran; at 65℃; for 16h; To the mixture of 450 mg <strong>[59702-07-7]1-<strong>[59702-07-7]methylpiperazin-2-one</strong></strong> (3.00 mmol) and 1.00 g K2C03 (7.24 mmol) in 5 mL TI-IF I mL 2-bromoethanol (14. Immol) was added and the mixture was stirred at 65°C for 16h. The mixture was cooled to room temperature, filtered andconcentrated under reduced pressure. The residue was purified via flash chromatography using DCM and MeOH to give 4-(2-hydroxyethyl)-1-methyl-piperazin-2-one.MS: (M+H) 159.4.
