YieldReaction ConditionsOperation in experiment
67% Dissolved in 3 ml of CH2Cl2 at room temperatureCompound 4 (1.00g, 2.70mmol) and Compound 5 (548.45mg, 2.35mmol) was successively added HATU (1.3g, 3.4mmol) and DIPEA (584mg, 4.5mmol).After stirring for 8 hours the volatiles by rotary evaporation, the residue was dissolved in EtOAc (25mL), washed with 1% HCl, saturated aqueous NaHCO3, washed with brine, dried over anhydrous Na2SO4. The dried solution was concentrated under reduced pressure and used in the next reaction without purification. The solid CH2Cl2 solution obtained above was added to HNEt2, and stirred at room temperature for 2 hours. After the volatile matter was removed by rotary distillation, a silica gel column was obtained to afford Compound 6 (657.94mg, 67%).
