Biological Activity

Description
CFDA-SE is a fluorescent dye capable of cell membrane penetration. It reacts with free amine groups in the cytoskeleton proteins inside the cell to form a fluorescent protein complex. Once inside the cell, CFDA-SE localizes to the cell membrane, cytoplasm, and nucleus, with the most intense fluorescence staining present in the nucleus[1].

In Vitro:

Cell Line
Concentration Treated Time Description References
rat hepatic oval cells (HOC) 5 μmol/L 10 minutes The positive rate of CFDA-SE labeled HOC was 90%, with no significant change in cell viabilities. Fluorescence intensity decreased progressively with cell division. PMC2665147
Newborn rabbit RPE cells 20 μM 1 minute To evaluate the effect of CFDA-SE on cell morphology and marker protein expression, results showed that CFDA-SE labeling did not affect cell morphology or marker protein expression. PMC2568003
pea chloroplasts 80 µM 20 minutes To evaluate the uptake and digestion ability of CFDA-SE in chloroplasts, the results showed that CFDA-SE was taken up by chloroplasts and digested by esterases, releasing high levels of fluorescence. PMC5723387

In Vivo:

Species
Animal Model
Administration Dosage Frequency Description References
Wistar rats fulminant hepatic failure (FHF) model intrahepatic injection Midazolam 9 mg/kg, Nitrous oxide 70%, Isoflurane 0.75% Single administration, lasting 2 hours or 6 hours HOC transplantation can improve liver function and increase the survival rate of recipients. Serum ALT, AST, and TBil levels decreased, while serum Alb levels increased after transplantation. PMC2665147
Hartley guinea pigs Spontaneous osteoarthritis model Intra-articular injection 5 × 10^6 cells per rat single injection To evaluate the migration, differentiation, and proliferation of CFDA-SE-labeled MSCs in an osteoarthritis model. Results showed partial cartilage repair in the HA-MSC group, and CFDA-SE-labeled cells demonstrated migration, differentiation, and proliferation in the HA-MSC group. PMC3392826
Mice Pregnant mouse model Intraductal pancreatic infusion Approximately 105 cells Single injection, observed for 2 months To assess the phenotype and fate of insulin-positive cells in pancreatic ducts, it was found that a small portion of CFDA-SE-labeled duct cells became insulin-positive, but they were not fully functional beta-cells, and these cells did not migrate into preexisting islets. Dissolving Methods
Please choose the appropriate dissolution scheme according to your animal administration guide.For the following dissolution schemes, clear stock solution should be prepared according to in vitro experiments, and then cosolvent should be added in turn:

in order to ensure the reliability of the experimental results, the clarified stock solution can be properly preserved according to the storage conditions; The working fluid for in vivo experiment is recommended to be prepared now and used on the same day;

The percentage shown in front of the following solvent refers to the volume ratio of the solvent in the final solution; If precipitation or precipitation occurs in the preparation process, it can be assisted by heating and/or ultrasound.
